Dense Nuclear Cataract Caused by the B-Crystallin S11R Point Mutation
نویسندگان
چکیده
RESULTS. A point mutation (A to C) of the B-crystallin gene, which results in the B-S11R mutant protein, was identified in this cataractous mouse line. Homozygous mutant mice developed dense nuclear cataracts associated with disrupted inner lens fiber cells. Immunohistochemistry data revealed -crystallin aggregates at the cell boundaries of inner mature fibers that lose actin filaments. Western blotting showed an increased degradation of crystallin proteins correlated with the nuclear cataract. ICP-OES confirmed a substantial elevation of calcium concentration in mutant lenses.

Connexin Mediated Cataract Prevention in Mice
Cataracts, named for any opacity in the ocular lens, remain the leading cause of vision loss in the world. Non-surgical methods for cataract prevention are still elusive. We have genetically tested whether enhanced lens gap junction communication, provided by increased α3 connexin (Cx46) proteins expressed from α8(Kiα3) knock-in alleles in Gja8tm1(Gja3)Tww mice, could prevent nuclear cataracts ...
متن کاملCataract-associated mutant E107A of human γD-crystallin shows increased attraction to α-crystallin and enhanced light scattering
Several point mutations in human γD-crystallin (HGD) are now known to be associated with cataract. So far, the in vitro studies of individual mutants of HGD alone have been sufficient in providing plausible molecular mechanisms for the associated cataract in vivo. Nearly all the mutant proteins in solution showed compromised solubility and enhanced light scattering due to altered homologous γ–γ...
متن کاملA novel deletion variant of gammaD-crystallin responsible for congenital nuclear cataract.
PURPOSE To investigate a novel deletion variant of gammaD-crystallin (CRYGD) identified in a Chinese family with nuclear congenital cataract. METHODS A Chinese family with five affected members diagnosed with nuclear cataract and four unaffected members were recruited for the mutational screening of 15 known candidate genes for autosomal dominant congenital cataract. Two-point linkage analysi...
متن کاملA novel mutation in CRYAB associated with autosomal dominant congenital nuclear cataract in a Chinese family
PURPOSE To identify the genetic defects associated with autosomal dominant congenital nuclear cataract in a Chinese family. METHODS Clinical data were collected, and the phenotypes of the affected members in this family were recorded by slit-lamp photography. Genomic DNA was isolated from peripheral blood. Mutations were screened in cataract-associated candidate genes through polymerase chain...
متن کاملA novel γD-crystallin mutation causes mild changes in protein properties but leads to congenital coralliform cataract
PURPOSE To identify the genetic lesions for congenital coralliform cataract. METHODS Two Chinese families with autosomal dominant coralliform cataract, 12 affected and 14 unaffected individuals, were recruited. Fifteen known genes associated with autosomal dominant congenital cataract were screened by two-point linkage analysis with gene based single nucleotide polymorphisms and microsatellit...
